The crash occurred at about 11:05 p.m. in the 3200-block of South DuSable Lake Shore Drive.
Police said the woman was driving southbound when the car hit the median and flipped over, landing upside down between the northbound and southbound lanes.
The woman was transported to the University of Chicago Medical Center, where she was pronounced dead, police said.
Two 29-year-old male passengers were transported to Northwestern Memorial Hospital in fair and good conditions, police said.
A 33-year-old woman who was a passenger was transported to a hospital as a precaution, police said.
Area Three detectives are investigating the crash.
